The effect of vitamin K2 on the whole organism

Osteocalcin is a protein dependent on vitamin K2, after synthesis it is in an inactive form. Vitamin K2 activates osteocalcin, which in its activated state is able to incorporate calcium into bones. Vitamin K2-MK7 is necessary for the synthesis of Matrix Gla Protein (MGP), which takes place in smooth muscle, blood vessel cells and cartilage. This protein is thought to be a strong inhibitor of blood vessel calcification. Vitamin K2-MK7 deficiency increases the level of non-carboxylated MGP, which is not functional, which leads to increased calcium deposition in blood vessels. Again, vitamin K2 is of great importance, because in addition to osteocalcin, it also activates vascular GMP, a protein that prevents the deposition of calcium in soft tissues. This explains the better structure and preservation of cartilage, as well as better joint function in people who take enough vitamin K2.

The influence of vitamin D on numerous functional systems of the body and the development of chronic pain in the field of physiatry competence

Vitamin D belongs to the group of liposoluble vitamins, although it is actually a hormone due to many characteristics such as chemical structure, synthesis in the body, regulation of synthesis, existence of receptors, pleiotropic effects. It belongs to the group of steroids, like sex hormones, mineralocorticoids and glucocorticoids. Like other hormones, it is synthesized from this provitamin D3 precursor, passes through the liver and kidney, where it is metabolized to its active form. It binds to its specific receptors and thus exerts its effects in the body. The knowledge that a significant number of people around the world have varying degrees of vitamin D deficiency stimulated interest in this issue and boosted research not only in the sphere of osteoporosis and bone metabolism, but also in many other areas with the aim of explaining the mechanism of the numerous pleiotropic effects of the D hormone and defining its necessary levels for manifesting pleiotropic effects. The greatest interest of the scientific public in this matter is directed in the direction of immunomodulatory effects and possible therapeutic effects in a large number of diseases with an immunological pathophysiological basis. Research is also being directed in relation to the connection between widespread chronic musculoskeletal pain and vitamin D deficiency, with great interest in clarifying the origin of vitamin D deficiency, as well as the possibilities of therapeutic influence on reducing chronic musculoskeletal pain. It is the duty of the physiatrist to take into account the possibility of vitamin D insufficiency affecting numerous functional systems, and to take timely therapeutic action. The teamwork of endocrinologists, rheumatologists and physiatrists is necessary for timely diagnosis and optimal treatment of the mentioned conditions.
